Ben - Certainly the .380 is superior to the 22 mag for self defense. But the "best" for your wife is whatever she feels more comfortable with. Let her try both a revolver and one or two of the excellent .380 semi-autos on the market. Check the Springfield

Ben, I would always choose a center fire round over a rim fire round. If you want a 380 revolver fine, but I would just get my wife a nice 38 special chambered revolver. Like one of these:--budsgunshop.com/catalog/product_info.php/products_id/26498/s%26w+
